The present study was performed to determine whether acetaminophen (paracetamol)
use after the measles-mumps-rubella vaccination could be associated with
autistic disorder. This case-control study used the results of an online
parental survey conducted from 16 July 2005 to 30 January 2006, consisting of 83
children with autistic disorder and 80 control children. Acetaminophen use after
measles-mumps-rubella vaccination was significantly associated with autistic
disorder when considering children 5 years of age or less (OR 6.11, 95% CI
1.42—26.3), after limiting cases to children with regression in development (OR
3.97, 95% CI 1.11—14.3), and when considering only children who had
post-vaccination sequelae (OR 8.23, 95% CI 1.56—43.3), adjusting for age,
gender, mother's ethnicity, and the presence of illness concurrent with
measles-mumps-rubella vaccination. Ibuprofen use after measles-mumps-rubella
vaccination was not associated with autistic disorder. This preliminary study
found that acetaminophen use after measles-mumps-rubella vaccination was
associated with autistic disorder.

Abstract
Circulating immunoglobulin E (IgE) is one of the characteristics of human allergic
diseases including allergic asthma. We recently showed that infection ofhuman B cells with
rhinovirus or measles virus could lead to the initial steps of IgE class switching.
Since many viral vaccines are live viruses, we speculated that live virus vaccines
may also induce IgE class switching in human B cells. To examine this possibility,
we selected the commonly used live attenuated measles mumps rubella (MMR) vaccine. Here,
we show that infection of a human IgM+ B cell line with MMR resulted in the expression of
germline transcript. In addition, infection of freshly prepared human PBLs with this
vaccine resulted in the expression of mature IgE mRNA transcript. Our data suggest that a
potential side effect of vaccination with live attenuated viruses may be an increase in
the expression of IgE. Copyright 2001 Academic Press.

Abstract: Histories obtained in 350 of 375 clinically confirmed cases of SSPE reported to a national registry showed that 292 patients had measles and 58 had no history of measles. Forty of the latter patients received live, attenuated measles virus vaccine. In patients with a history of measles, measles illness occurred before age 2 years in 46%, and a mean of 7.0 years before onset of SSPE. In contrast, there was no relationship of SSPE with age at vaccination in 35 of 40 patients historically associated with measles vaccine and SSPE occurred a mean of 3.3 years after vaccination. Based on estimated national measles morbidity data and national measles vaccine distribution data, the risk of SSPE following measles vaccination (0.5 to 1.1 cases/106) appears to be less than the risk following measles (5.2 to 9.7 cases/106 ). Because live measles vaccine is highly effective in preventing measles illness and a high proportion of children in the US have received measles vaccine, these data are consistent with the observed downward trend in SSPE incidence since 1969.

Immunization of mice with a number of live virus vaccines
(poliovaccine, smallpox vaccine, measles vaccine) given consecutively at 14-day intervals
resulted in increased frequency of chromosomal aberrations in bone marrow cells of the
animals after the completion of the entire vaccination course (14 and 30 days after the
last vaccination). Measles vaccine and, particularly, smallpox vaccine exert a significant
harmful effect on the karyotype of the bone marrow cells. The effect on the chromosomes of
the vaccines given consecutively differs somewhat from the individual effect of each of
them.
